Can 7-AAD be fixed?
7-AAD is widely used for live/dead cell discrimination. It also can be used in fixed and permeabilized cells for cell cycle profiling by DNA content analysis using flow cytometry. 7-AAD intercalates selectively at GC-rich regions of DNA, making it dye useful for chromosome banding studies.
Is 7-AAD cell permeable?
Recent work by Idziorek et al. (1995) also revealed that following initiation of apoptotic cascade plasma membrane becomes selectively permeable to small, cationic molecules such as cyanine dyes. At the same time it remains impermeable to larger cations such as PI or 7-AAD.
What is 7-AAD used for?
7-AAD has been used for cell cycle analysis by flow cytometry. 7-AAD also binds selectively to GC regions of DNA yielding a distinct banding pattern in polytene chromosomes and chromatin for use in chromosome banding studies.
What color is 7-AAD?

Is 7-AAD toxic to cells?
I didn’t observe any toxicity due to 7AAD in the samples and results are just fine everytime. I will recommend using them together to avoid the over complications in protocol. 7AAD binds well in Annexin binding buffer also. You can wash the dyes off by PBS once incubation is completed.
How does 7-AAD stain dead cells?
Several different fluorochromes can be used to stain non-viable cells including 7-amino actinomycin D (7-AAD). 7-AAD is a membrane impermeant dye that is generally excluded from viable cells. It binds to double stranded DNA by intercalating between base pairs in G-C-rich regions.
Can you fix cells after Annexin V staining?
Yes, cells can be fixed with formaldehyde after staining. Because Annexin V staining is dependent on calcium, all buffers used for washing and fixation should contain 1.25 mM CaCl2. Fixation may increase the background signal from Ethidium Homodimer III.
How does annexin V detect apoptosis?
Annexin V was shown to interact strongly and specifically with PS and can be used to detect apoptosis by targeting for the loss of plasma membrane asymmetry. Labeled annexin V can be applied both in flow cytometry and in light microscopy in both vital and fixed material by using appropriate protocols.
What is anti annexin?
Anti-annexin V antibodies (aANVAs) are detected in various diseases like rheumatoid arthritis, systemic lupus erythematosus and anti-phospholipid antibody syndrome. The study of ANV in Acute Myocardial Infarction (AMI) might shed light on hypercoagulability mechanisms in the pathogenesis of acute coronary syndromes.
Does annexin V stain necrotic cells?
Annexin V can also stain necrotic cells because these cells have ruptured membranes that permit Annexin V to access the entire plasma membrane.
What does annexin V measure?
Annexin V used for measuring apoptosis in the early events of cellular cytotoxicity. Cytometry.
